I have an Apple Iic with ROM 4x and a Floppy Emu connected to the SmartPort and emulating a 5.25 drive. Booting ProDOS 2.4.3 as an external drive works just fine. Booting ProDOS 2.5.0a8 causes a crash to the monitor or a hang with "PRODOS..." appearing on screen. I used Copy II+ to create a physical disk with 2.5.0a8 and it boots just fine from the internal 5.25 drive.
